Mario is obviously the first character, but the game establishes at the beginning that Lakitu is the one controlling your camera. Sure, you could write this off as a trick to familiarize the player with the camera controls (a new concept at the time), but the game sticks to the conceit of Lakitu being the one with the camera by having his reflection shown in the mirror room pretty well into the game.
Since you control the camera, and since Lakitu is the camera/camera holder, this means that Lakitu is a playable character by virtue of the fact that you control him. You simply do it at the same time that you control Mario. Maybe this was the true meaning of "L is real." Lakitu, not Luigi. =P
